Overview

About this product

Retatrutide (LY3437943) is a synthetic peptide designed to activate three receptors: the glucose-dependent insulinotropic polypeptide receptor (GIPR), the glucagon-like peptide-1 receptor (GLP-1R) and the glucagon receptor (GCGR). That makes it a reference compound for research into multi-receptor agonism, receptor bias and how the three pathways interact. Because three receptors can contribute to one observation, useful designs include single-receptor agonists or receptor-specific cell lines as comparators. Available in 10 mg and 20 mg vials, each supplied as a lyophilized powder in a sealed vial. What the research shows Retatrutide has been studied in randomised human trials. In the phase 3 TRIUMPH-1 trial, published in 2026, 2,339 adults with obesity and without diabetes were randomised to retatrutide or placebo for 80 weeks; mean body weight changed by -25.0% in the highest-dose group and by -3.9% with placebo, and the most common adverse events were gastrointestinal. In the 40-week phase 3 TRANSCEND-T2D-1 trial of 537 adults with type 2 diabetes, glycated haemoglobin changed by -1.69% to -1.94% across the three retatrutide groups and by -0.81% with placebo, and no severe hypoglycaemia was reported. Purity and identity are stated on the batch certificate, published on this page once it is on file.

Counted on PubMed on 2026-10-20. A score shows how much published research covers an area: 1 for fewer than 5 papers, 2 under 20, 3 under 100, 4 under 500 and 5 from 500, plus half a point when the area includes randomised controlled trials and a full point from 10 of them (5 at most). It measures the amount of research, not an effect. Each count links to its PubMed search.

The molecule

About this compound

Retatrutide is a synthetic peptide of 39 amino acids. In the published literature it is also identified as LY3437943, and it is described as engaging the GLP-1, GIP and glucagon receptors. That is the published description of the molecule. Peptune sells it as a 10 mg vial and a 20 mg vial. Those figures are the contents of each vial. They are not a dose.
