Overview

About this product

Tirzepatide is a 39-residue synthetic peptide that activates two incretin receptors, the GIP receptor and the GLP-1 receptor. It is acylated with a C20 fatty diacid chain that binds albumin, which lengthens its half-life in research models. Its receptor pharmacology, including a stronger preference for the GIP receptor than for GLP-1R, is well characterised, which makes it a common reference point in incretin studies. It is often compared with single-receptor GLP-1 agonists and with the triple agonist retatrutide. Supplied as a lyophilized powder in a sealed vial. What the research shows Tirzepatide has been studied in large randomised human trials. In the phase 3 SURMOUNT-1 trial, published in 2022, 2,539 adults with obesity, or overweight with at least one weight-related complication, and without diabetes were assigned to tirzepatide or placebo for 72 weeks; mean weight change was -15.0% to -20.9% across the three tirzepatide groups and -3.1% with placebo. In the 40-week SURPASS-2 trial, published in 2021, 1,879 patients with type 2 diabetes were assigned to tirzepatide or semaglutide; glycated haemoglobin changed by -2.01 to -2.30 percentage points with tirzepatide and -1.86 with semaglutide. In both trials the most common adverse events were gastrointestinal. Sources: Jastreboff et al., New England Journal of Medicine, 2022, PMID 35658024. Purity and identity are stated on the batch certificate, published on this page once it is on file.

Counted on PubMed on 2026-10-20. A score shows how much published research covers an area: 1 for fewer than 5 papers, 2 under 20, 3 under 100, 4 under 500 and 5 from 500, plus half a point when the area includes randomised controlled trials and a full point from 10 of them (5 at most). It measures the amount of research, not an effect. Each count links to its PubMed search.

The molecule

About this compound

Tirzepatide is a synthetic peptide of 39 amino acids. In the published literature it is also identified as LY3298176, and it is described as engaging the GIP and GLP-1 receptors. A C20 fatty diacid is part of that published structure. It comes in 10 mg, 20 mg and 30 mg vials, and the figure on the label is the contents of the vial.
